    A macro picture of the eye isn’t so easy, as you’ve found. A tip would be to try continuous focus (servo mode on Canon), putting the focus point on the edge of the iris. Shoot in burst mode and you should get some keepers. If you can hold still long enough, go single shot, but take plenty of exposures. You need the focus point on the edge of the iris to let the af lock on. Give it a try, you’ll see what I mean. Another tip is to use auto iso and dispense with the flash. Good luck.
